Summary
The activity of an enzyme is usually measured by quantifying a compound produced by the action of the enzyme on a substrate dissolved or dispersed in water. However, in the case of an enzyme that acts on a water-insoluble substrate such as cellulose, the substrate cannot be dissolved in water, so the above method cannot be easily applied. The present invention is a new method for measuring the activity of an enzyme whose substrate is insoluble or sparingly soluble in water and has the following constitution.
1) A predetermined amount of enzyme is arranged on a part of the surface of a gel body in which a part of the dispersoid is a substrate of the enzyme.
2) The volume of the dent formed on the surface of the gel body by the action of the enzyme is measured, and the activity of the enzyme is calculated based on the measurement result and the predetermined amount of the enzyme.
In addition to the method of measuring the volume of the depression, the method of measuring the change in the light transmittance of the gel body due to the formation of the depression and the method of measuring the change of the light reflectance of the surface of the gel body due to the formation of the depression are also possible to calculate the enzyme activity.
